How does this option work? If I select this while a clip is playing does that mean the clip won't play again within this VGHD session or does it mean that the clip will NEVER play again? And if it's the latter, how can I get it back? (Like if I had clicked the option by accident).

It means that the clip will never play again. However, you can reset all the shows for a girl to play again by going to My Collection in Options and clicking on the model and then click on "Reset Disabled Clips".
Also I requested that Totem add a prompt for the user in case they accidently click on this option for a clip. I'm not sure if the feature has been added in this latest version yet or not since I haven't tried it.
